摘要
目的研究内关穴位注射香丹注射液预处理对大鼠心肌缺血炎性因子白介素-6(IL-6)、白介素-10(IL-10)的影响,明确内关穴位注射香丹注射液预处理对心肌的保护作用,进而揭示穴位注射预处理对心肌保护作用的机制,为防治冠心病提供新途径。方法本研究以SD大鼠为受试对象,心肌缺血前给予穴位注射预处理14d,然后通过腹腔注射异丙肾上腺素(ISO)造成心肌缺血模型,采用酶联免疫吸附法(ELISA)检测IL-6、IL-10。结果①促炎因子IL-6在大鼠心肌缺血后表达增多,内关组、曲池组及药物对照组预处理后均可以降低其表达,内关组的这一作用明显优于曲池组及药物对照组;②抗炎因子IL-10在心肌缺血后含量较正常组有增多趋势,内关组、药物对照组及曲池组预处理后可以升高其含量,内关组的这一作用明显优于曲池组及药物对照组;③内关组、药物对照组和曲池组的IL-6/IL-10与模型组比较差异均有统计学意义(P<0.01),内关组IL-6/IL-10与曲池组和药物对照组比较差异均有统计学意义(P<0.01),曲池组与药物对照组比较则无明显差异。结论内关穴位注射香丹注射液预处理,可以升高血清IL-10含量,降低IL-6含量,这可能是对心肌缺血大鼠心肌具有保护作用的机制之一。
Objective To explore a new way about preventing the cure of ischemic heart disease,we study the effects of pretreatment by acupoint injection of Xiangdan parenteral solution at Neiguan on IL-6,IL-10 in rats with acute myocardial ischemia. Methods 14 days before myocardial ischemia,SD rats were performed acupoint injec- tion of Xiangdan parenteral solution at Neiguan acupoint injection. ISO was us'ed by peritoneal injection to establish the rat model of acute myocardial ischemia. Then we observed the changes of IL-6 and IL-10 in serum of each group by ELISA. Results (1) The level of IL-6 was increased in the model group compared to the normal group,and decreased in the Neiguan group,drug control group and Quchi group compared to the model group. (2) The level of IL-10 was increased in the Neiguan group,drug control group and Quchi group compared to the normal group and model group. (3)Compared to the model group, the level of IL-6/IL-10 was statistically decreased in Neiguan group, drug control group and Quchi group. Compared to the drug control group and Quchi group,the level of IL-6/IL-10 was significantly lower in Neiguan group. Conclusion The pretreatment by acupoint injection of Xiangdan paren- teral solution at Neiguan acupoint can increase the level of IL-10, but decrease the level of IL-6, which maybe one of the mechanism for pretecting cardiac muscle in rats with acute myocardial ischemia.
