摘要
目的建立家兔血浆中甘露醇含量的测定方法。方法家兔血浆样品沉淀蛋白后,加入高碘酸钠使之氧化成为甲醛,再加入Nash试剂显色,在412nm处采用比色法进行测定。结果用本法测定的血浆中的甘露醇在0.50~6.00mg/ml范围内线性关系良好,相关系数r为0.9987;其平均加样回收率为97.59%,RSD为2.51%。结论采用本法测定血浆中甘露醇含量的精密度和重复性良好,并且操作简单易行。
Objective To establish a method of determination of plasma mannnitol content in rabbits. Methods After plasma samples from rabbits were deproteinized, mannitol was oxidated with sodium periodate and further reacted with Nasb reagent,and then its content was detected at a wavelength of 412 nm by colorimetry. Results Plasma mannitol level showed a good linearity within the range of 0.50 - 6. 00 mg/ml. The correlation coefficient was 0. 9978, the average recovery was 97.59% ,and RSD was 2.51%. Conclusion This method for measuring rabbit plasma mannitol is of good precision and repeatability. It is feasible and easy to operate.
