摘要
目的探讨端粒酶和端粒酶逆转录酶(hTERT)在胃黏膜癌变过程中的作用及其过度表达与胃癌恶性病理生物学行为之间的关系。方法采用聚合酶链反应端粒酶重复序列扩增酶联免疫吸附测定法和免疫组化链菌素亲物蛋白过氧化酶法分别检测端粒酶和TERT在胃癌组织和癌旁非癌组织中的表达,计算阳性细胞的百分率。结果65例胃癌组织中,端粒酶和hTERT蛋白阳性表达率分别为86.2%和81.5%,显著高于相应癌旁非癌组织(P<0.01)。端粒酶活性表达与胃癌组织的淋巴结转移及PTNM分期显著相关(P<0.05),hTERT的表达和肿瘤浸润深度、淋巴结转移和PTNM分期显著相关(P<0.05)。结论检测端粒酶和hTERT可以帮助判断胃癌的发生、浸润、转移和临床分期。
Objective To explore the roles and relationships of telomerase and telomerase reverse transcriptase (TERT) in carciomgenesis of gastric cancer. Methods The telomerase activity and TERT protein in gastric cancer tissues and matched normal adjacent tissues were determined by PCR TRAP-ELISA assay and SP immunohistochemical staining assay,and percentage of positve rate was evaluated. Results Positive rate of telomerase and TERT protein in 65 gastric cancer tissues were, significantly higher than these of matched normal adjacent tissues( 86.2% and 81.5%, P〈0.01). Significant correlation was found between either lymph node metastasis or PTNM stage and telomerase activity in gastric cancer. The TERT overexpression correlated with tumor invasion, tumor infiltration depth, lymph node metastasis, and PTNM classification. Conclusion The detection of telomerase and TERT protein will help evaluate carcinogenesis, infiltration, metastasis and clinical stage of gastric cancer.
