摘要
目的:对粘性放线菌5519Ⅰ型菌毛进行分离和部分特性研究。方法:采用超声粉碎方法从粘放菌5519细胞壁上分离Ⅰ型菌毛,然后经160000×g24h高速离心,硫酸铵沉淀和葡聚糖凝胶G100过柱后取得纯化菌毛,并经电镜和SDS-PAGE电泳检查。结果:粘放菌5519Ⅰ型菌毛大部分分子量为53573.4u,少量为37699.8u。经电镜检查证实为Ⅰ型菌毛。结论:通过超声,结合高速离心。
Aim: We described the isolation and partial characterization of type l fimbriae from Actinomyces viscosus 5519 cell walls. Methods: Surface type 1 fimbriae, Viscosus 5519 were released from cell walls by ultrasonic . The removed fimbriae were sedimented for 24h at 160,000g, then were purified by repeated precipitation with ammonium sulfate, and collected in fractions obtained at the void volume of an sephadex G100. This preparation was further examined by electron microscopy and SDS-PAGE electrophoresis. Results: The results showed that most of the molecular masses of type 1 fimbriae was 53573.4u, small parts was 37699.8u. Electron microscopy of this molecular masses revealed fimbriae. Conclusions: Purified type 1 fimbriae of A viscosus 5519 can be obtained by ultrasonic combine with sediment, precipitation and chromatography of sephadex G100.
