摘要
目的探讨肝细胞癌(HCC)中细胞凋亡情况及其与P53和Bcl2蛋白表达的关系.方法细胞凋亡采用原位细胞凋亡检测法P53和Bcl2蛋白表达采用免疫组化方法.细胞凋亡与P53,Bcl2蛋白表达的关系采用双标记法进行检测.结果HCC70例,癌组织和癌周组织中细胞凋亡指数(1000个细胞中)分别为36±20和106±36.Bcl2和突变型P53蛋白检出率分别为229%和429%.Bcl2主要表达在癌周组织中,抑制肝细胞凋亡;而P53则仅表达在癌组织,抑制癌细胞凋亡.双染色显示细胞凋亡与P53或Bcl2不同时在一个细胞中表达.结论HCC癌细胞凋亡减弱.突变型P53蛋白是癌细胞凋亡减弱的主要原因,而Bcl2则可能在维持HCC的肝脏功能方面具有重要作用.
AIM To investigate apoptosis in HCC, and its relation to the expression of Bcl 2 and P53 protein. METHODS In situ method was used to detect apoptosis. IHC was used to detect the expression of Bcl 2 and P53 proteins. Double staining was used to demonstrate the relationship between apoptosis and the expression of Bcl 2 or P53 protein. RESULTS Apoptotic indexes were 3 63±1 96 and 10 63±3 64 (per 1000 cells) respectively in cancerous tissues and pericancerous tissues of 70 HCC cases. Positive rates of the expressions of Bcl 2 and mutant type P53 proteins were 22 9% and 42 9% respectively. Bcl 2 proteins mainly expressed in pericancerous tissues, and down regulated apoptosis of hepatocyte. Mutant type P53 proteins, however, mainly expressed in cancerous tissues, and down regulated apoptosis of cancer cells. CONCLUSION Apoptosis of HCC cancer cells was weakened. The overexpressions of mutant type P53 proteins might be the main cause of it. Bcl 2 proteins might play an important role in maintaining the liver function of HCC patients.
