豆制品中总异黄酮含量的紫外分光光度测定法 被引量:2

Detection of Total Isoflavone in Soybean Products by Ultraviolet Spectrophotometry

摘要 目的建立豆制品中总异黄酮含量的紫外分光光度检测法。方法将烘干的豆制品研成粉末,取粉末样品置于索氏抽提器中,用乙醚脱脂5h后,用80%的甲醇溶液以25∶1的液固比热回流5h,用紫外分光光度法测定滤液总异黄酮含量。结果方法的最大吸收波长为254nm,线性范围为0~100mg/L,相关系数r=0.9979,平均回收率为97.82%,相对标准偏差1.46%,RSD为1.7%。结论该方法简单,线性好,使用的试剂毒性小且廉价易得,不需特殊精密检测仪器,适用于基层单位。 [ Objective ] To establish the ultraviolet spectrophotometry for the detection of total isoflavone in soybean products. [ Methods ] The drying soybean products were triturated. The powder samples were defated by the diethyl ether for 5 hours, and returned by the 80% methyl alcohol solutions with the liquid-solid ratio of 25:1 for 5 hours. The content of the total isoflavone in filtrate was determined by ultraviolet spectrophotometry. [ Results ] The maximum absorption wavelength of the method was 254 nm, the liner range was 0 - 100 mg/L, the correlation coefficient of standard curve was 0. 9979, the average recovery rate was 97.82%, the relative standard deviation was 2.3% and RSD was 1.7%. [ Conclusion] The method haS advantages of simple operation, good linearity, cheaper reagents with little toxic, and it does not need the special precision instrument. The method is suitable for basic level unit.
