摘要
目的优化PCR程序,尝试用降落PCR扩增人类常染色体STRD15S128。方法用普通PCR和降落PCR扩增人类常染色体STRD15S128。结果普通PCR扩增产物有非特异带,降落PCR无非特异带。结论降落PCR简化了普通PCR中摸索最适退火温度的过程,可以一步找到人类常染色体STRD15S128的退火温度,是一种高效率的PCR方法。
Objective To optimize PCR program and try to amplify human autosomal STR D15S128 by touchdown PCR（TD-PCR）. Methods We used both common PCR and TD-PCR to amplify human autosomal STR D15S128. Results Ordinary amplification showed non-specificity band and TD-PCR showed no non-specificity band. Conclusion Compared with ordinary PCR,TD-PCR is simple and effective.
