款冬花配方颗粒制备工艺优化及质量控制 被引量:5

摘要 目的优化款冬花配方颗粒的制备工艺,建立指标成分芦丁的含量测定方法。方法以正交实验法对款冬花配方水提工艺的影响因素进行考察,优选工艺条件;用HPLC法测定芦丁含量,检测波长为360nm,流动相为甲醇-0.1%磷酸水溶液(50:50)。结果优选提取工艺为:10倍量水,回流提取3次,1.5h/次。芦丁提取率为85.53%,出膏率为25.95%;芦丁在1~12.5μg(r=0.9998)范围内呈良好线性关系。平均回收率为99.07%(n=5)。结论款冬花配方颗粒的制备工艺稳定、可行;评价指标可靠、合理。 Objective To optimize the extraction process for the preparation of Tussilago farfara formula granules and to establish an HPLC method for the determination of rutin in the samples. Methods The optimum water extraction process was selected by the orthogonal design and the content of turin in formula granules was analyzed by HPLC method. The HPLC assay was performed by using MeOH -0.1% phosphoric acid (50: 50) . The detection wavelength was set at 360 nm. Results The optimum extraction conditions were to reflux for 3 times with 10 folds of water and each time for 1. 5 h. The extraction rate of ratin was 85. 53% , and the dried extraction yield was 25.95%. The peak area had a good linearity with the concentration of rutin and the linear range was 1 - 12.5 μg( r =0.999 8 ). The average recovery was 99.07% ( n = 5 ). Conclusion The preparation process of Tussilago farfara formula granules is stable and feasible. The HPLC method for quality control is accurate and suitable.
