摘要
目的探讨葶苈生脉方治疗压力超负荷致慢性心衰(Chronic heart failure,CHF)大鼠的作用机理。方法采用腹主动脉缩窄法建立慢性心衰大鼠模型,随机分为假手术组、模型组、葶苈生脉方高剂量组(简称高剂量组)、葶苈生脉方低剂量组(简称低剂量组)、心宝丸对照组(简称心宝丸组),各组给予相应处理因素4周后,采用放免法检测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6),Motic6.0图象分析系统分析心肌胶原容积分数(CVF)。结果模型组与假手术组比较TNF-α、IL-6表达显著增强(P<0.01),CVF明显升高;各治疗组与模型组比较,TNF-α、IL-6表达显著减弱(P<0.01),CVF明显降低,其中高剂量组明显低于低剂量组和心宝组(P>0.01)。结论葶苈生脉方可能通过抑制炎性细胞因子的表达,调节心肌胶原代谢,从而阻止或逆转心室重构,改善心功能,达到治疗CHF的目的。
Objective To explore the effects of Tinglishengmaifang on heart failure rat model induced by pressure overload. Methods The model with chronic heart failure was established by suprarenal aortic constriction operation, then was divided into 5 groups at random : Sham operated group, model control group, Tinglishengmaifang high - dose group ( referred to as high - dose group) , Tingfishengmaifang low -dose group( referred to as low -dose group) and Xinbaowan control group group( Referred to as Xinbaowan group). Four weeks after treatment, the contents of TNF-α and IL- 6 were determined by radiation- immunization method, and motic 6.0 Digital Image Analysis System was used to analyze myocardial collagen volume fraction (CVF). Resuits Compared with the normal group, the contents of TNF - , IL - 6 in model group were significantly inereased, CVF was significantly enhanced(P 〈 0.01 ) ; Compared treatment group with the model group,TNF- α, IL- 6 were significantly weakened, CVF was significantly reduced in treatment groups. Conclusion Tilishengmaifang can inhibit the expression of inflammatory factor, regulate collagen metabolism and reverse ventricular remodele, improving heart function, so to achieve the purpose of treating CHF.
