Synthesis of Rabeprazole Sodium

摘要 雷贝拉唑钠(rabeprazole sodium,1),化学名为2-[[[4-(3-甲氧基丙氧基)-3-甲基-2-吡啶基]-甲基]亚磺酰基]-1H-苯并咪唑钠盐,由日本卫材开发,1998年首次在日本上市,临床用于治疗十二指肠溃疡与胃溃疡、反流性食管炎等消化系统疾病。本品起效快、抑酸效果好,可24h持续抑酸、缓解症状。 Rabeprazole sodium, an antiulcer drug, was prepared from 2-chloromethyl-4-(3-methoxypropoxy)- 3-methylpyridine hydrochloride and 2-mercapto-1H-benzimidazole via condensation, oxidation with sodium hypochlorite and salt formation with an overall yield of about 71%.
