摘要
目的探讨不同分化程度的胃癌细胞凋亡情况和全反式维甲酸对其影响。方法以人胃癌细胞系低分化的MKN45和高分化的MKN28细胞作为研究对象,以不同浓度的全反式维甲酸干预其72小时后,以原位DNA断段切口标记法和DNA琼脂糖电泳检测细胞凋亡情况。结果两种细胞系的凋亡率均低于5%,全反式维甲酸干预后凋亡指数上升,且可能与药物浓度有关。结论全反式维甲酸可诱导胃癌细胞凋亡。
Objective To induce gastric cancer cell apoptosis with folic acid and to evaluate its anticarcinogentic mechanism. Methods The ability of folic acid to induce the onset of gastric cancer cell apoptosis was investigated by using human gastric cancer cell lines MKN 45 and MKN 28 and the DNA fragmentation was studied in situ by TdT mediated dUTP biotin nick end labeling and DNA agarose gel electrophoresis. Results The rates of apoptosis in both poorly differentiated MKN 45 and well differentiated MKN 28 cell lines were less than 5%. Apoptosis could be induced by administration of all trans retinoic acid and apoptosis indices of MKN 45 and MKN 28 cells were related to dosage of the drugs. Conclusion It is concluded that induction of gastric cancer cell apoptosis by folic acid may play a role in the anticarcinogenesis, so it may possibly be used as a therapeutic agent in the treatment of human gastric cancer.
