摘要
目的:细胞遗传学研究发现,鼻咽癌染色体7q32-qter常有缺失。为进一步证实鼻咽癌在这个染色体区域的遗传变化。方法:选择位于7q313~36的13个微卫星标记对24例鼻咽癌进行等位基因缺失分析。结果:79%(19/24)的鼻咽癌至少在一个位点存在等位基因丢失,高频缺失位点是D7S495(46%),D7S500(45%),D7S631(30%)和D7S514(35%)。依据缺失图谱分析,一个较小的共同缺失区域可能存在于7q32的D7S495和D7S500之间,其遗传距离约为96cM。且这个小区域的缺失与鼻咽癌的临床分期无显著相关性(P>005)。结论:这些结果提示,鼻咽癌在7q32的D7S495与D7S500之间可能存在与鼻咽癌相关的抑瘤基因。
Objective:To better define genetic alterations of chromosome 7q32-qter,because the deletions of chromosome 7q32-qter are commonly observed in nasopharyngeal carcinoma (NPC). Methods: PCR-assays for the analysis of 13 microsatellite markers mapped to chromosome 7q31 3~36 were performed on 24 NPC patients. Results:Allelic imbalance and loss of hetreozygosity were found in 19 of the 24 tumor specimens (79%) at least one locus.Higher frequent allelic losses were identified at four loci,which were D7S495(46%),D7S500(45%),D7S631(30%) and D7S514(35%).A smaller commonly deleted region was supposed to be present between D7S495 and D7S631 at 7q32 and their genetic distances might be within 9.6 centimorgan.The LOH/deletion on chromosome 7q32 did not correlate with the clinical staging of NPC( P >0 05).Conclusion:A putative tumor suppressor gene may be present at a smaller region within 10cM on chromosome 7q32, which may be important for cancer initiation and early development in nasopharyngeal carcinoma.
