盐酸多奈哌齐对侧脑室注射Aβ25-35大鼠海马脑源性神经营养因子表达的影响 被引量:4

摘要 目的观察盐酸多奈哌齐(DN)对侧脑室注射Aβ25-35大鼠海马脑源性神经营养因子(BDNF)表达的影响。方法3月龄Wistar大鼠24只随机分为对照租、干预组、模型组,每组8只,向大鼠侧脑室立体定向注射Aβ25-35建立AD动物模型,然后干预组予以腹腔注射DN,对照组与模型组腹腔注射等量0.9%氯化钠溶液,最后用免疫印迹法检测大鼠海马BDNF的表达。结果免疫印迹法测定结果显示模型组大鼠海马BDNF的表达量较对照组下降,差异有统计学意义(P<0.01),干预组大鼠BDNF的表达量较模型组增加,差异亦有统计学意义(P<0.05)。结论侧脑室注射Aβ25-35的大鼠其海马BDNF的表达较对照组明显下降,干预组与模型组相比BDNF的表达增加,盐酸多奈哌齐可能会增加AD模型大鼠海马BDNF表达。 Objective To explore the effect of donepezil (DN) on the expression of brain - derived neurotrophic fac- tors (BDNF) in hippocampus of rats induced by Aβ25 - 35. Methods Forty - eight Wistar rats were divided randomly into groups control, DN and model, 8 in each. Ad animal models were established by stereotactically injecting Aβ25 - 35 into the lateral ventricle. DN group were injected with DN for 4 weeks. The expression of BDNF in hippocampus was also determined by Western blotting. Results The expression of BDNF decreased in model group when compared with control group, (P 〈 O. 01 ), and increased in treatment group as compared with model group ( P 〈 0. 05 ). Conclusion BDBF expression decreases signifi- cantly in rats injected with Aβ25 - 35, and increases in intervention group as compared with model group. Donepezil hydrochlo- ride may increase BDNF expression in hippocampus of AD model rats.
