鱼体中硝基苯残留量测定方法的改进研究

Study on the improvement of testing method of nitrobenzene residue in the fish body

摘要 为了监测自然水体中鱼体内硝基苯的残留量,对硝基苯的环境污染程度做出准确的评估,试验优化了鱼体中硝基苯残留量的测定方法。在比较索氏提取法和水蒸气蒸馏提取法的基础上,采用改进的水蒸气蒸馏提取法,结合苯反萃取,最后用气相色谱法检测,最终形成了改进后的鱼体中硝基苯残留量的检测方法。与索氏提取法和水蒸气蒸馏提取法相比,该方法所需时间短、有机溶剂消耗量少、操作简便、回收率高。在质量比3.0~500.0 μg·kg^-1内线性关系良好,线性方程为Y=0.161 3X-0.527 8,r=0.999 9。方法检出限为3 μg·kg^-1,加标回收率为75%~110%,相对标准偏差为3.64%~6.13%。结果证明该方法适用于鱼体中硝基苯残留量的检测。 The paper is aimed to present an improved testing method for monitoring the nitrobenzene residue level in the fish body in natural water bodies and assessing the environmental pollution by the nitrobenzene. For our research purpose, we have done a comparative study with various extraction solvents, stripping solvents, purification methods and concentration methods by choosing acetone as extraction solvent, and benzene as stripping solvent. Based on the comparison of soxhlet extraction and traditional steam distillation extraction methods, we have developed a novel method for testing the nitrobenzene residue in the fish body. The testing procedure can be shown as follows: when the acetone was added to the fish sample and benzene was extracted by using the homogenized, improved extraction method, or by the liquid-liquid extraction method, or by NaCl solution method, the residue level can be tested and measured by the gas chromatography. With our improved novel method, a steam generating bottle could first of all be added to the extraction procedure, which helps to get rid of bumping and increase the extraction efficiency. Secondly, it was also possible to get rid of the purification procedure, which may get rid of the risk of residue recovery with the time and mainpower saved. In so doing, the chief parameters of the new method, such as time consumption, demand of organic solvent volume, simplicity of working procedure and recovery prove all to be better than the other currently used methods. Within the linear range of 3.0 - 500.0μg·kg^-1, the detection limit turns out to be 3.0μg·kg^-1, with recovery rates ranging from 75% to 110%, and with the relative standard deviation standing between 3.64% and 6.13% . The results of application of the innovated method show that the proposed method is simple, fast, sensitive, eco-friendly, and, therefore, is feasible for testing nitrobenzene residues in the fish body.
