胰岛素抵抗大鼠血压升高的血管机制 被引量:9

摘要 目的:探讨胰岛素抵抗在高血压发病中的作用和血管机制。方法:复制胰岛素抵抗大鼠模型,采用血管功能实验方法观察其血管收缩及舒张功能的变化以及胰岛素对血管舒缩功能的影响。结果:(1)高糖喂养一个月能引起正常大鼠出现胰岛素抵抗和血压升高;(2)胰岛素抵抗大鼠血管对苯肾上腺素的收缩反应增强,自动舒张功能减弱;(3)胰岛素能抑制血管的收缩反应,促进血管的舒张和直接舒张血管,但这些作用在IRH大鼠中均减弱或消失。结论:胰岛素抵抗是发生高血压病的因素之一。 AIM and METHOD:To investigate the effect of insulin resistance on blood pressure and its possible vascular mechanisms,we developed a rat model of insulin resistant hypertension (IRH)by feeding with high glucose diet.Alteration of contractile response and automatic relaxation were studiet in isolated aortic,mesenteric and renal arteries.RESULTS:(1)High glucose feeding induced insulin resistance and hypertension in rats.Blood pressure was increased by 5.3~8 0 kPa in IRH rats.(2)The vasoconstrictive responses to phenylephrine were higher in IRH rats.The automatic relaxations were significantly slower in IRH rats than in controls.The time to half relaxation(T 50 )of aortic,mesenteric and renal arteries from IRH rats were (160±28)s,(130±24)s and(120±32)s respectively,while that from the controls were(120±20)s,(90±15)s and (86±13)s( P <0 01).(3)Insulin inhibited vasoconstriction,potentiated vasorelaxation,and directly caused relaxation.All these functions were reduced or abolished in IRH rats.CONCLUSION:That insulin resistance may induce hypertension.Alteration of vascular responses and the abnormalities of regulatory effect of insulin on vasocular responses may be involved in the mechanisms.
