摘要
目的:研究帕金森病(PD)黑质多巴胺(DA)神经元的死亡机制。方法:用脱氧核糖核酸(DNA)标记法,借助荧光显微镜观察多巴胺对PC12细胞凋亡的影响。结果:DA可诱发PC12细胞凋亡,适中浓度时(0.45mmol/L)PC12细胞凋亡数最多(凋亡率为48.7%±6.3%);抗氧化剂N乙酰半胱氨酸(NAC)可部分阻止DA诱发的PC12细胞凋亡(P<0.01)。结论:凋亡参与了PD的病变过程。
Objectives:To study the mechanism of dopaminergic neuronal death in substantia nigra of Parkinson's diseaes(PD). Methods:The effect of dopamine(DA) on apoptotic PC12 cell were examined by fluorescence microscopy using the DNA end-labeling methods. Results:DA may induce apoptosis in PC 12 cell,moderate concentrations of DA(0.45mmol/L4) giving maximal number of apoptotic cell (rate of apoptosis:48.7%±6.3%);and the antioxidant N-acetyl-cysteine(NAC) partially prevent DA-induced apoptosis in PC12 cells(P>0.01). Conclusion:Apoptosis may be involved in the pathogenesis of PD, and treatment with proper antioxidants amy be useful in PD therapy.
