摘要
目的研究尼莫地平对脑缺血再灌注大鼠脑组织差异蛋白的影响,进一步揭示尼莫地平的脑保护作用及抗损伤修复机制。方法将12只大鼠随机分为脑缺血再灌注模型组和尼莫地平组(每组6只),分别于造模前3d灌胃给予等量生理盐水和尼莫地平20 mg·kg^(-1),每日1次,d3于手术前1h给药。线栓法致缺血再灌注24h后,分别提取脑蛋白质,2-DE电泳分离蛋白质,应用ImageMaster 2D V2002.1软件比较分析,找出差异蛋白,酶切消化后进行肽指纹图谱分析,数据检索,初步鉴定差异蛋白质。结果与模型组比较,从尼莫地平组找到了25个差异表达蛋白斑点(10个低表达,15个高表达),鉴定其中6个蛋白质。结论尼莫地平对脑缺血再灌注的保护作用是多个蛋白共同参与的结果。
AIM To study the effects of nimodipine on the different proteins in brain tissue of rats with focal cerebral ischemia-reperfusion (CIR) and to learn the mechanism of nerve-protecting effects of nimodipine. METHODS Twelve rats were divided randomly into CIR model group and nimodipine group (6 for each). The nimodipine group was given nimodipine 20 mg.kg^-1 by gastrointestinal once daily, while the CIR model group was given normal saline for 3 days before reperfusion. After CIR of 24 h by the suture-occluded method, brain tissue protein was exercted, 2-DE was applied to separate proteins, and Image-Master 2D (V2002.1) software was used to find different proteins. Then peptident mass fingerprint spectra (PMF) analysis was done after enzyme digestion to initially identify the different proteins through database search. RESULTS CIR model, there were 25 different proteins found in the nimodipine group, 15 proteins up- Compared to regulated and 10 proteins down-regulated, and 6 spots of them were identified and characterized. CONCLUSION The protective effect of nimodipine on CIR is the result of the participation of a number of proteins.
