摘要
利用等电聚焦(IEF)电泳和不连续醋酸尿素聚丙烯酰胺凝胶电泳(NAU-PAGE)对玉米籽粒贮藏蛋白的等电点(pI)和在F1中的遗传表现以及贮藏蛋白在胚和胚乳中的分布进行研究,结果表明:(1)玉米籽粒贮藏蛋白的pI在3.5-8.45范围内,可分离的有39条左右,60%左右的蛋白质属酸性,pI分布在3.50-6.85范围内,40%左右属中性偏碱,pI在6.85-8.45范围内。(2)籽粒贮藏蛋白在F1代呈共显性遗传,没有偏亲现象。所有被测组合在F1代均有互补带,这是区别自交系与杂交种有效的生化依据。(3)蛋白质在胚中百分含量较高,即浓度较高,胚乳中较低。
Isoelectric focusing (IEF) electrophoresis and non coherent acetic acid urea polyacrylamid gel electrophoresis (NAU PAGE) were used in this paper to study maize seed storage protein components and their characteristics.The results showed as follows:(1) Isoelectric pionts of maize seed storage proteins ditributed from 3.50 to 8.45.About thirty nine bands of storage proteins were clearly seperated by IEF electrophoresis,about 60% storgae protein bands of being distinguished 39 protein bands with pI from 3.50 to 6.85,the others with pI from 6.86 to 8.45.(2) Seed storage protein components of double parents all emerged in their hybrid seeds and existed at a higher percent rate in the embryo than in the endosperm.(3) All hybrids analysed in this paper had protein complementary bands of double parents,which was a good character to distinguis inbred seeds from their hybrid seeds.
