家蚕斑纹限性品种SC_1的RAPD差异DNA片段的克隆及其酶切图谱分析

摘要 利用RAPD技术对家蚕限性普斑品种SC1的雌雄体分别进行PCR扩增通过120种引物的筛选,得到1个雌体特异的RAPDDNA片段将该片段克隆到pUC19质粒,井进行了限制性内切酶图谱分析. The sex chromosome formula of the silkworm, Bombyx mori L are designated as ZW for the female and ZZ for the male. In order to identify random amplified polymorphic DNA(RAPD) drived from the T(W;Z)+ p chromosome, the normal larval marking gene + p was translocated to the W chromosome. Cenodric DNAs from the females and males of the SCI strain [Dp(Z; W) + p, p/p] were used as templates for PCR. Approximately 120 primers (10 - mers) were screened for polymorphisms between male and female. One specific DNAfragment was found unique in female, but absent in male. The fragment was cloned into plasmid pUC19 and its restriction map was analyzed. This female - specific RAPD was located on the T(W; Z) + p chromosome and would be useful for the analysis of the W chromosome.
