摘要
本文对抗枯萎病性不同的12个陆地棉品种胚轴外植体进行了培养、比较,并就枯萎菌毒素对其愈伤组织诱导的影响进行了研究。发现,(1)12个品种胚轴外植体在 Ls+IAA 2mg/L+KT 1mg/L 培养基上,均能产生胚性和非胚性两类不同性质的愈伤组织;(2)枯萎菌毒素提取液经高压灭菌后,仍具有较强的毒害活力;(3)枯萎菌病毒素提取液对陆地棉胚轴外植体具有很强的毒害作用,从而抑制了胚轴外植体愈伤组织的产生,且随浓度增加而作用愈强;(4)抗病性不同的品种对枯萎菌毒素的反应情况不同,高抗品种受毒害较轻,且能产生较多的愈伤组织,而高感品种受害较重,且很难形成愈伤组织;(5)采用逐代提高毒素浓度的培养方法,对愈伤组织无性系进行继代筛选,获得了少量抗高浓度毒素的愈伤组织无性系。这些结果为进一步通过组织细胞培养,获得抗病植株及试管快速鉴定抗病品种等工作奠定了基础。
Twelve upland cotton cultivars of different resistance to fusarium wilt disease were cultured and the effect of toxin extract of cotton fusarium wilt pathogen (Fusarium oxysporum f.vasinfectum (ATK) Sayol & Hans) on callus induction was studied.Two kinds of vigor calli were obtained from the hypocotyls of all cultivars.One is fragile and light green or light grey colorea,while the other is hard and white or dark green colored.The toxin extract was resistant to high temperature,and it possessed a very strong toxicity to exlants of cotton hypocotyls.The callus induction of cotton was inhibited by the toxin,and the higher the concentration of toxin contained in the medium,the higher the toxicity affected to explants.Explants from the cultivar which has higher resistance to the desesse in whole plant level produce more calli than those from lower resistance one.This result may be used to assay the resistance of cotton cultivars to fusarium wilt desease.Through progressively increasing the concentration of toxin extract contained in the medium,some callus of resistant to higher concentration of toxin extract had been obtained.
