摘要
目的探讨限制能量摄入对缺血一再灌注老年大鼠心功能的影响。方法 18个月龄SD大鼠经过限制能量摄入(给予正常饮食量的60%)8个月,建立限制能量摄入动物模型。取离体心脏行Langendorff装置灌流,停灌复灌模拟全心缺血一再灌注,记录其心功能变化,留取冠脉流出液测定LDH含量,灌注末取心脏测定心梗死面积。结果限制热量组与正常饮食组比较,体重、心重明显低于正常饮食组,血糖水平明显降低。缺血一再灌注后两组大鼠心功能均受损,但限制热量组心功能受损程度较轻。限制热量组冠脉流出液中LDH释放量减少、心肌梗死面积减少。结论限制能量摄入可改善老年大鼠缺血一再灌注的心功能。
Objective To investigate the effect of caloric restreiction on global myocardial ischemia reperfusion injury in old rats. Methods Eighteen -month -old Sprague -Dawley (SD) rats received caloric restriction(60% of regular intake)for 8 months. Rats in caloric restriction group and control group were killed at the end of the evaluation of physical constitution. The hearts were quickly removed,and mounted on Langendorff apparatus, and perfused with Krebs - Henseleit ( K - H) solution. Except for control group, all hearts were followed by 30 min no - flow ischemia and 30 min reperfusion. A computer - based electrophysiological recorder system was used to measure changes of heart rate(beats/min) , systolic and diastolic pressure(mmHg) , aortic flow (ml/min). The infarct size was tested by triphenyltetrazolium chloride. The release of lactate dehydrogenase(LDH) in the coronary effluent was determined with LDH kit. Results The body weight and heart weight didn' t increase significantly in caloric resiriction group,as compared to normal food group. The glouse concentration decreased significantly in caloric resiriction group. The heart function was subjected to injury in both groups. But the caloric resiriction rats' heart function injury was lighter than the normal food group. The release of lactate dehydrogenase (LDH) in the coronary effluent and the infarct size was decreased in the caloric resiriction group. Conclusion Caloric resiriction can improve the heart function of the old rats for global myocardial ischemia reperfusion injury.
