摘要
目的通过免疫组化法观察复方浙贝颗粒联合阿霉素对 K562/A02移植瘤细胞耐药相关酶 GST、Topo-Ⅱ表达影响。方法将 K562/A02细胞接种 BALB/c—nu 裸小鼠腋前皮下构建 MDR 移植瘤动物模型,给予复方浙贝颗粒灌胃联合阿霉素腹腔注射,实验结束后处死小鼠,剥离肿瘤,将瘤块组织制成切片,通过免疫组化结合 Image ProPlus 6.0免疫组化彩色图像分析,观察复方浙贝颗粒联合阿霉素对 K562/A02移植瘤细胞耐药相关酶 GST、Topo-Ⅱ表达的影响。结果与阿霉素组比较,CZBG各剂量组移植瘤组织细胞相关酶 GSH、Topo-Ⅱ的 IOD 值明显降低(p<0.05)。结论 CZBG 能协同阿霉素抑制 K562/A02移植瘤组织细胞耐药相关酶 GSH,Topo Ⅱ的高表达。
Objective To investigate the effects of Compound Zhe Bei granule (CZBG) combined with doxorubicin on the expression of GST and Topo-Ⅱ in K562/A02 cell line multidrug resistance tumor xenografts in mice. Methods Tumor xenografts model was established by injecting the multidrug resistance cell line K562/A02 in the axillary flank of BALB/c - nu - nu mice. Drug - comgbination of CZBG intragastric administration and doxorubicin intraperitoneal injection ( i. p. ) was given to the BALB/c - nu nude mice. The tumor xenografts were made into slice after the dissection, and the expression of GST and Topo-Ⅱ in K562/A02 tumor xenografts in mice was investigated by immunohistochemical technique. The integral optical density (IOD) of GST and Topo-Ⅱ in K562/A02 tumor xenografts was measured by Image ProPlus 6.0. Results Compared with the single treatment of doxorubicin i. p group,the combination of the doxo- rubicin and CZBG with dosage classified by three types( high, middle, low) can decrease IOD of GSH and Topo-Ⅱ in K562/A02 tumor xenografts with statistical significance(P〈0.05). Conclusion The combination of CZBG and doxorubicin can decrease the expression of GST and Topo-Ⅱ in K562/A02 multidrug resistance tumor xenografis in mice.
