甜菜碱对脂肪组织释放游离脂肪酸和抗氧化能力的影响 被引量:4

Effect of Betaine on Free Fatty Acids Release and Anti-oxidant Capacity from Adipose Tissue

摘要 目的观察甜菜碱对大鼠离体脂肪组织释放游离脂肪酸(Free fatty acid,FFA)和抗氧化能力的影响,并探讨其作用机制。方法将大鼠离体脂肪组织置于37℃Krebs液中温育,分别加入不同浓度的甜菜碱及各阻断剂,观察其对FFA和丙二醛(MDA)、总抗氧化能力(T-AOC)的影响。结果甜菜碱可促进脂肪组织释放FFA(P=0.000),并存在剂量相关效应(r=0.909,P=0.000);甜菜碱可降低脂肪组织MDA含量,提高T-AOC;心得安、异搏定和SQ22,536(腺苷酸环化酶抑制剂)可不同程度的阻断上述作用。结论甜菜碱可促进离体脂肪组织释放FFA,提高其抗氧化能力。此作用可能部分经由脂肪细胞的肾上腺素能β受体、L型Ca2+通道和腺苷酸环化酶所介导。 Objective The effect of betaine on free fatty acid(FFA) release and anti-oxidant capacity from isolated adipose tissue in rats and its mechanism were studied. Methods Isolated rat tissues were placed into Krebs solution to incubate at 37℃ and were respectively added into the betaine of different concentration and blockers, the FFA values and malondialdehyde (MDA) and tatal anti-oxidant capacity(T-AOC) content from isolated adipose tissue were observed. Results Betaine could promote the FFA release (P = 0. 001 ;r = 0. 909, P = 0. 000)dose-dependently. It could reduce MDA and raise T-AOC from isolated adipose tissue which can be blocked partly by propranolol, varapamil or SQ22,536 (adenylyl cyclase inhibitor). Conclusion Betaine can enhance the catabolism of adipose tissue in vitro by increasing the release of FFA and improving its antioxidant capacity. The effect may be mediated partly by β3-adrenergic receptor, varapamil-sensitive Ca2+ channel L and adenylyl cyclase(AC).
