神经生长因子对人卵巢组织激素分泌和KL mRNA表达的影响

摘要 目的:研究神经生长因子对体外培养的人卵巢组织雌孕激素分泌及KL mRNA表达的影响。方法:将10例人卵巢皮质切成薄片分成5组:未培养组、基础培养组、NGF组、抗NGF组及同型抗体对照组,培养结束后行雌孕激素测定及RT-PCR检测KL mRNA表达变化。结果:NGF组的KL mRNA表达0.91±0.30,略高于基础培养组的0.83±0.32,差异无统计学意义(P>0.05)。NGF组5个不同时间点的雌激素水平均高于其余各组,各组孕激素水平比较无统计学差异。结论:NGF能促进体外培养的人卵巢组织雌激素的分泌。 Objective: To investigate the effects of nerve growth factor (NGF) on hormone secretion of human ovarian tissue and expression of KL mRNA in vitro. Methods: Ovary cortex of 10 patients were cut into ovarian pieces and divided into 5 groups: non - culture group, basic culture group, NGF group, anti - NGF antibody group and homotype antibody group, after cultivation, the levels of estradiol and progestogen were detected, and the expression of KL mRNA was measured by RT - PCR. Results : There was no difference in KL mRNA expression between NGF group (0. 91 ±0. 30) and basic culture group (0. 83±0. 32) (P 〉0. 05) . The levels of estradiol in 5 different time points in NGF group were significantly higher than those in the other groups, but there was no significant difference in progestogen level among the five groups. Conclusion: NGF can promote the secretion of estradiol of human ovarian tissues in vitro culture.
