摘要
利用摇瓶分批补料培养模式考察了补料基质对红曲霉JR的菌体生长以及红曲色素色阶的影响。考察了碳源补加种类(葡萄糖、蔗糖和麦芽糖)、蛋白胨补加浓度对红曲霉JR发酵的影响,结果表明:碳源的补加能显著促进菌体生长及红曲色素色阶的提高,葡萄糖是最佳的补加碳源;补加蛋白胨也显著促进红曲霉JR的菌体生长。在红曲霉JR摇瓶分批补料培养模式下,第48h、72h、96h补加2.0g/100mL发酵液的葡萄糖和0.5g/100mL发酵液的蛋白胨,红曲色素色阶达到331.6±8.7U/mL,显著高于摇瓶分批培养的色阶(105.7±5.8 U/mL)。
The effects of feeding medium on cell growth and production of Monascus pigments by Monascus purpureus JR were investigated in the shaking flask. Firstly, the effects of various carbon sources (glucose, sucrose and maltose) feeding on the fermentation of M. purpureus JR were determined. The results indicated that feeding the three carbon sources could significantly enhance the cell growth and monascus pigment content. Feeding glucose (2.0 g/100ml ) and peptone (0.5g/100mL) at 48, 72 and 96 h could greatly stimulate the cell growth and pigments production. Under these conditions, the content of monascus pigment could reach 331.6±8.7 U/ml, which was significantly higher than that without feeding carbon and nitrogen (105.7±5.8 U/ml).
