天然脑活素对复制性衰老细胞端粒酶活性影响的实验研究 被引量:1

摘要 目的探讨天然脑活素对复制性衰老人胚肺二倍体成纤维细胞(MRC-5)端粒酶活性的影响,以阐明其延缓衰老的作用及分子机制。方法镜下观察细胞形态变化,绘制细胞生长曲线,MTT法检测细胞增殖活度,TRAP-ELISA方法检测细胞端粒酶活性的变化。结果天然脑活素孵育的MRC-5细胞老龄化不明显,细胞增殖能力较空白老龄对照组明显增加(P<0.05);天然脑活素处理的细胞端粒酶活性较空白老龄对照组明显上调(P<0.05)。结论天然脑活素可能通过促进细胞增殖,激活细胞端粒酶的表达,进而有效地延缓细胞的复制性衰老。 Objective To explore the effect of natural cerebrolysin on the activity of telomerase of replicativc senescence human embryonic lung diploid fibroblasts( MRC -5) so as the explain its action on delaying aging and molecular mechanism. Methods To observe the changes of cellular morphology microscopically, draw the cell growth curve,detect the activity of cell proliferation by MTT,and detect the changes in the activity of telomerase by TRAP - ELISA method. Results The aging was not apparent for MRC - 5 cells in- cubated with natural cerebrolysin. The cell proliferation was intensified apparently compared wilh blank aged control group(P 〈0.05). Telomerase activity processed by natural cerebrolysin was up -regulated obviously compared with that in blank aged control group( P 〈 0.05 ). Conclusion Natural cerebrolysin delays effectively cell replicative senescence by accelerating cell proliferation and activating the expression of telomerase probably.
