摘要
植物凝集素可引导植物-微生物之间的相互识别,对植物根际促生细菌的定殖起着重要作用。采用石油醚和丙酮脱脂、甲酸抽提、硫酸铵盐析、热处理、甲壳素亲和层析的方法分离纯化麦胚凝集素(WGA),从小麦胚中纯化WGA的纯化倍数为115.9,血凝活力为6.8 mg/L,产率为0.566 mg/g。为检测WGA与小麦根际细菌的亲和性识别作用,依据Marshall法对纯化的WGA进行异硫氰酸荧光素(FITC)标记;用标记的WGA对小麦根际细菌染色,26株小麦根际促生细菌中23株具有FITC-WGA染色阳性。表明这些FITC-WGA染色阳性的菌株和WGA具有亲和性识别作用,揭示WGA可能影响定殖在小麦根际的促生细菌。
Wheat germ agglutinin(WGA) can induct the recognition between plant and microorganisms,and help the colonization of plant growth-promoting rhizobacteria(PGPR) on plant roots.Purification process of WGA was conducted by using means of petroleum ether and acetone degreasing,formic acid extraction,ammonium sulfate salt out,heat treatment and chitin affinitive chromatography.Results show the purity of the produced WGA increases by a factor of 115.9,the hemo-agglutination activity of the product is 6.8 mg/L,and the yield is 0.566mg/g wheat germ.To detect affinity between WGA and PGPRs,the purified WGA was labeled with FITC by the method of Marshall,and the labeled WGA was applied to wheat PGPR strains.Results show 23 of 26 PGPR strains are stained,indicating there is affinity between the staining positive strains and WGA,and WGA is likely to influence colonization of PGPRs in wheat rhizosphere.
