摘要 核糖体展示技术是20世纪90年代建立并发展起来的一种完全在体外进行的分子筛选与进化技术,避免了体内展示技术的缺陷,不受细胞转染与基因表达等因素影响,已经成功用于筛选与靶分子特异结合的高亲和力多肽、抗体和酶等。本文主要概述了核糖体展示技术的基本原理、具体步骤、技术改进及应用现状。 Ribosome display technology,which was established and developed in 1990s,is a method for completely in vitro selection and evolution of functional molecules.It circumvents many drawbacks of in vivo systems,and is not influenced by cell transfection and gene expression.The technology has already been successfully used to optimize the affinity and specificity of peptides,antibodies and enzymes in binding target molecules.This review summarized the principle,procedure,improvement and application of ribosome display.
