摘要 目的以中国广东省汉族人群为研究对象,探讨Tolls样受体4(TLR4)等位基因Asp299Gly多态性与肺炎易感性、肺炎严重程度的关系。方法采用病例对照研究和PCR产物测序的方法,检查60例肺炎患者和40名健康体检者的TLR4等位基因Asp299Gly的分布与肺炎严重程度的关系。结果在肺炎患者和健康体检者中,TLR4等位基因Asp299Gly基因型所有个体均为野生型纯合子,Asp299Gly野生型与突变型在肺炎患者与健康个体,或重症肺炎与非重症肺炎患者中的分布无差异。结论TLR4等位基因Asp299Gly多态性与肺炎易感性、肺炎严重程度相关性不明确。 Objective To determine the relationship between Toll-like receptor 4 (TLR4) gene Asp299Gly polymorphism and human susceptibility to pneumonia or seversity of pneumonia. Methods Sixty patients with pneumonia and 40 healthy controls residing in Guangzhou, China were enrolled. Blood samples were obtained from all subjects. DNA samples were extracted following the instructions of Blood Genome DNA Extraction Kit. The sequence containing Asp299Gly allele (A→G) was amplified. The PCR products were confirmed by agarose gel electrophoresis and sequenced. In addition, clinical data of all pneumonia pa- tients were documented. Results The Asp299Gly allele of the TLR4 gene was not identified in all 100 specimens of the Cantonese. The prevalence of TLR4 genotypes was essentially the same between pneumonia patients and control subjects. Furthermore, there was also no association with the subgroup of pneumonia patients (severe pneumonia or non-severe pneumonia). Conclusions Our findings indicate that TLR4 gene Asp299Gly polymorphism dose not influence the prevalence of pneumonia and its relationship with severity of pneumonia is unclear.
