摘要
为了提高牛巴贝斯虫核酸探针的检测性能,制备了地高辛标记的牛巴贝斯虫C15A核酸探针。该探针检测牛巴贝斯虫DNA的灵敏度为32pg,检测探针DNA的灵敏度为0.1pg;检测其他对照血液原虫(双芽巴贝斯虫、边缘无浆体、瑟氏泰勒虫、伊氏锥虫、卵圆巴贝斯虫)和牛血细胞的DNA,均未出现非特异性反应。与光敏生物素标记的牛巴贝斯虫C15A核酸探针比较,光敏生物素标记探针能检出10%带虫血12.5μL,而地高辛标记探针能检出10%带虫血0.015μL,且杂交背景浅,显色深。
A dig labelled C51A probe was prepared to improve the specifility and sensitivity of C51A probe for detection of Babesia bovis. The dig labelled probe was shown to be able to detect 32 pg of Babesia bovis DNA and 0.1 pg of probe DNA and did not react with the DNAs of Babesia bigemina, A.marginale, T.sergenti, Trypanosoma evansi or bovine blood cell. In comparison with the photo biotin labelled one, the dig labelled C51A probe enable to detect 0.015 μL instead of 12.5 μL of 10% infected blood with a lower hybridization background and darker blotting.
