摘要
目的通过观察高胆固醇血症患者B族Ⅰ型清道夫受体表达的变化,探讨B族Ⅰ型清道夫受体在动脉粥样硬化发生中的作用及其表达变化的机制。方法随机选取健康对照60例和高胆固醇血症患者80例,肘静脉取血,分离血清、血浆并提取血小板,放射免疫法检测血浆血管紧张素Ⅱ水平,硝酸还原酶法检测血清一氧化氮含量,实时定量聚合酶链反应和免疫印迹法分别检测血小板B族Ⅰ型清道夫受体的mRNA和蛋白表达水平。结果高脂组的血浆血管紧张素Ⅱ水平和血清一氧化氮水平均较对照组明显升高(P<0.01)。高脂组血小板B族Ⅰ型清道夫受体的mRNA和蛋白表达均较对照组明显降低(P<0.05和0.01)。高脂组的血管紧张素Ⅱ水平与B族Ⅰ型清道夫受体的表达呈显著负相关(r=-0.488,P<0.05)。结论高胆固醇血症患者血小板B族Ⅰ型清道夫受体表达的降低与血管紧张素Ⅱ的刺激有关。
Aim To investigate the effects of scavenger receptor class B type Ⅰ ( SR-B Ⅰ ) in atherosclerosis by observing the change of SR-B IⅠexpression. Methods Eighty patients with hypercholesterolemia and sixty healthy persons were investigated as hyperlipidemia group and control group respectively. Blood samples were extracted from ulnar vein. Blood serum, plasma and platelets were extracted and stored at a refrigerator of - 80℃. The level of angiotensin Ⅱ( Ang Ⅱ) in plasma and nitric oxide ( NO ) in serum were detected by the method of radioimmunity and nitrate reductase respectively. The expression of SR-B Ⅰ mRNA and protein on platelet were assessed by real-time polymerase chain reaction ( RT-PCR ) and Western Blotting respectively. Results The plasma Ang Ⅱ and serum NO of hyperlipidemia group were higher than that of control group (P 〈 0.01 ). The expression of SR-B Ⅰ mRNA and protein on platelets were lower in hyperlipidemia group than those of control group ( P 〈 0.05 and 0.01 ). The expression of SR-B Ⅰ protein was inversely correlated with the plasma Ang Ⅱ ( r = - 0.488, P 〈 0.05 ) in patients with hypereholesterolemia. Conclusion The expression of SR-B Ⅰ decreases and the decrease of SR-B Ⅰ is related to Ang Ⅱ.
