胰岛素样生长因子-1对体外培养大鼠髓核细胞增生活性及代谢的影响 被引量:1

Effect of Insulin - like Growth Factor - 1 on the Proliferative Activity and Metabolize of the Rat Nucleus Pulposus Cells in vitro.

摘要 目的探讨不同浓度的胰岛素生长因子(IGF-1)对体外培养大鼠椎间盘髓核细胞分裂增生及细胞代谢的影响。方法体外单层培养大鼠腰椎髓核细胞,对照组培养液为含20%胎牛血清的DMEM,实验组为在培养液中加入IGF-1使其浓度分别为0.1μg/ml,1μg/ml,10μg/ml,100μg/ml在其作用3、5、7天,分别观测细胞形态,免疫组化检测Ⅱ型胶原,MTT法绘制生长曲线。结果 HE、甲苯胺蓝以及免疫组化染色显示实验组细胞量明显多于对照组,MTT法显示吸光度值,第3天时4组与其余组有差别P<0.05;5天时对照组与1组、3组、4组分别有差别P<0.05;7天时1、2组与3、4、5组均有差别P<0.05,1、2组无差别,3、5组与4组有差别P<0.05,3、5组无差别。结论髓核细胞在体外培养过程中,IGF-1能有效促进髓核细胞的增生,且随其浓度不断增加,促进增生作用显著增加,细胞代谢与因子浓度呈线性关系。 Objective To explore the effect of different concentrations of the insulin growth factor - 1 ( IGF - 1 ) on cell division proliferation and metabolism of rats nucleus pulposus cells in vitro. Methods Rats lumbar vertebra nucleus pulposus cell are cultured in monolayer in vitro. Culture fluid of control group is DMEM containing 20% fetal calf serum, and of experimental group mixed IGF - 1, make the concentration respectively of 0. 1 ng/ml, 1 ng/ml, 10ng/ml, 100ng/ml. Respectively observed cellular morphology, detected Ⅱ collagen by immunohistoehemistry, drawn growth curvature by MTT method in 3,5,7 day. Results The number of cells in experimental group significantly more than control group' s detected by HE, Toluidine blue and immunohistrochemistry stained method. Photometric quantities shows, by MTT method. The difference between group 4 and other group in 3 day, control group and group 1.3,4 in 5 day,group 1,2 and group 3,4,5 ,group 3.5 and group 4 in 7 day is respectively significant ( P 〈 0.05 ). but there' s no difference between group 1 and group 2 in 7 day. Conclusion Nucleus pulposus cells in vitro process, the insulin growth factor (IGF - 1 ) can promote effectively the nucleus pulposus cell proliferation, and with the increasing concentration significantly increased proliferation, promote cells metabolism, linear relationship with concentration.
