摘要
目的:研究补阳还五汤及其有效组分和三七总皂苷(total Panax notoginseng saponins,TSPN)含药血浆对血小板衍生生长因子(platelet-derived growth factor,PDGF)诱导的血管平滑肌细胞(vascular smooth musclecell,VSMC)增殖的影响。方法:采用血浆药理学方法及 PDGF—BB 诱导 VSMC 增殖模型。以甲基噻唑基四唑(methyl thiazolyl tet-razolium,MTT)法检测空白血浆对 VSMC 生长的影响和含药血浆对 PDGF—BB 诱导的 VSMC 增殖的半数抑制浓度,确定补阳还五汤及其有效组分和三七总皂苷用药浓度。MTT 法和流式细胞术检测补阳还五汤及有效组分和 TSPN、阿托伐他汀对 VSMC 增殖活性及细胞周期的影响。结果:PDGF—BB 刺激后,VSMC 增殖活性显著增强,G_0/G_1期细胞数减少,G_2/M 期和 S 期细胞数增多。与PDGF-BB 刺激组比较,补阳还五汤、有效组分生物碱和苷、TSPN、阿托伐他汀等含药血浆均可抑制 PDGF—BB 诱导的 VSMC 增殖,增加 G_0/G_1期细胞数,减少 G_2/M 期和 S 期细胞数。结论:补阳还五汤及其有效组分和 TSPN 均可抑制 PDGF—BB 诱导的 VSMC增殖。生物碱和苷可能为补阳还五汤中抗 VSMC 增殖的主要药效物质,补阳还五汤及其有效组分生物碱和苷具有与阿托伐他汀、TSPN相似的抗 VSMC 增殖的作用。
Objective: To explore the effects of plasma containing Buyang Huanwu Decoction (BYHWD), a compound of traditional Chinese medicine, and its effective components (alkaloids and glycosides) and total Panax notoginseng saponins (TSPN) on vascular smooth muscle cell (VSMC) proliferation induced by platelet-derived growth factor (PDGF) in vitro.
Methods: VSMC proliferation was induced by incubating VSMCs with PDGF-BB. Effects of different concentrations of blank plasma on the VSMC proliferation and 50% inhibitory concentrations of plasma containing BYHWD, its alkaloids and glycosides, TSPN and atorvastatin were detected by using methyl thiazolyl tetrazolium (MTT) method to determine the concentrations of plasma containing drugs for the following experiments. Then, MTT and flow cytometry methods were used to detect the effects of plasma containing BYHWD, its alkaloids and glycosides, TSPN and atorvastatin on VSMC proliferative activity and cell cycles.
Results: VSMC proliferative activity was enhanced obviously after PDGF-BB stimulation. The cell number in G0/G1 phase was decreased and the cell quantity in G2/M and S phases was increased following PDGF-BB stimulation. Compared with PDGF-BB stimulation group, VSMC proliferation induced by PDGF-BB in plasma containing BYHWD, its alkaloids and glycosides, TSPN and atorvastatin groups was inhibited. In all plasma containing drug components groups, the cell number of G0/G1 phase was higher and the cell quantity of G2/M and S phases was lower than those in the PDGF-BB stimulation group.
Conclusion: BYHWD and its alkaloids and glycosides have similar roles of resisting VSMC proliferation to TSPN and atorvastatin. Alkaloids and glycosides may be the major effective substances of anti-VSMC proliferation in BYHWD.
