定喘汤对支气管哮喘小鼠肺组织STAT1、STAT6及外周血γ-干扰素的影响 被引量:2

Effects of Dingchuan Decoction on expression of STAT1 ,STAT6 in lungs of asthmatic mice and interferon-γ in peripheral blood plasma

摘要 目的观察定喘汤对卵蛋白(OVA)致敏的支气管哮喘(哮喘)小鼠模型肺组织中信号转导和转录激活因子1(STAT1)、信号转导和转录激活因子6(STAT6)、外周血γ-干扰素(IFN-γ)的影响,并探讨其作用机制。方法雄性BALB/c小鼠随机分为5组。OVA腹腔注射和雾化吸入诱发哮喘发作后予药物治疗(对照组除外)。逆转录-聚合酶链反应(RT-PCR)法半定量测定肺组织中STAT1、STAT6 mRNA的表达水平,酶联免疫吸附试验(ELISA)检测外周血中IFN-γ的浓度,并常规做病理切片。结果与哮喘组(1.71、1.67、141.40)对照,定喘汤治疗组STAT1(0.72)、STAT6(0.68)明显降低(P<0.01),IFN-γ(375.99)明显升高(P<0.01)。结论定喘汤通过抑制STAT1、STAT6的表达及刺激IFN-γ生成来减轻哮喘时的气道黏膜炎症。 Objective To investigate the effects of Dingchuan Decoction on STAT1, STAT6 and interferon-γ(IFN-γ) in ovalbumin challenged asthmatic mice and to explore the possible mechanism to bronchial asthma(asthma). Methods BALB/c male mice were randomly divided into five groups. The mice were treated with drugs after sensitized and challenged except those in the normal group. One part of lung sample was carried through HE staining, others were employed to detect the expression of mRNA for STAT1 and STAT6 by RT-PCR. The concentration of IFN-γ in the peripheral blood were detected by ELISA. Results Compared with those of asthmatic group, the expression of STAT1, STAT6 mRNA in Dingchuan Dection group were significantly lower (P 〈 0.01 ) and the concentrations of IFN-γ was significantly higher( P 〈 0.01 ). Conclusions Dingchuan Decoction can significantly inhibit the mRNA expression of STAT1 and STAT6 in pulmonary tissue, decrease the concentration of IFN-γ of peripheral blood plasma, relieve asthmatic airway inflammation.
