摘要
目的研究仿生酶解法提取蜈蚣的工艺条件。方法以水解度为指标,单因素考察了仿生酶解法提取蜈蚣的工艺条件;以APTT法、纤维蛋白原平板法为考察指标,对仿生酶解法提取蜈蚣的工艺进行验证。结果仿生酶解法凝血时间长,抗血栓效果明显。酶解的条件为:蜈蚣先用37℃人工胃液加入4.0%(酶底比)胃蛋白酶酶解4h,残渣转入37℃人工肠液加入5%(酶底比)胰蛋白酶酶解4h。结论仿生酶解法适于提取蜈蚣抗凝血和溶栓的有效部分。
Objective To study the technology condition of extracting Scolopendra by bionic enzymatic method. Methods The technology condition of extracting Scolopendra by bionic enzymatic method was observed by single factor observation taken the hydrolysis degree as the index, and the extractive technique was verified by using the methods of active partial thromboplastin time (APTF) and fibrinogen plate. Results The coagulation time was longer and anti-thrombosis effect was more significant in the bionic enzymatic method. The best enzymatic condition was as follows:Scolopendra was hydrolyzed in artificial gastric juice (37℃ ) and pepsin ( ratio of pepsin to substrate was 4.0% ) for 4 hours, and then the residue was hydrolyzed in artificial intestinal juice (37 ℃ ) and trypsin ( ratio of pepsin to substrate was 5% ) for 4 hours. Conclusion The bionic enzymatic method can be used in the extraction of effective components of anti-coagulation and thrombolysis from Scolopendra.
