Screening and Identification of Laccase-producing Wood-rot Fungi and Characterization of Its Enzyme

摘要 综合菌体培养方式和培养基组成对菌株产漆酶性能的影响,对68株木腐菌进行了筛选,获得了1株适合液态发酵产漆酶的漏斗多孔菌A08,酶学性质研究表明,漆酶反应最适温度为60-65℃,最适pH为5.5,60℃时酶活半衰期大于45min,20℃酶活性在pH3.0-9.0稳定,漆酶活性受Cl-、Ca2+、Fe2+抑制,Mn2+激活,K+、Mg2+、Zn2+、Cu2+等对酶活性没有显著影响。 Based on the effects of culture method and medium composition upon productivity of laccase from Wood-rot Fungi, a strain with relative higher lacease-producing ability in liquid culture was screened from 68 strains of wood-rot fungi, and identified as Polyporus arcularius. Studying on enzymatic properties revealed that the optimum temperature and pH of the lacease reacted with guaiaeol were 60 - 65 ℃ and pH 5.5, respectively. The half-life of the laccase was more than 45 min at 60℃. The enzyme activity was stable within pH range of 3.0 -9.0 under 20℃ , was strongly inhibited by Fe^2+ , and moderately by Cl^- , Ca^2+ , while enhanced by Mn^2+ Wheres other ions like K^+ , Mg^2+ , Zn^2+ , Cu^2+ had no evident effects on it within the range of experimental concentration.
