摘要
目的:研究瘦素对大鼠背根神经节(DRG)神经元中Bcl-2和Pax蛋白表达的影响。方法:用不同浓度(1、10和100nmol/L)的瘦素处理体外培养大鼠DRG神经元;采用免疫印迹技术,检测神经元中Bcl-2和Bax蛋白的表达变化。结果:10和100nmol/L的瘦素能促进DRG神经元中Bcl-2蛋白的表达;瘦素对Pax蛋白表达没有影响。结论:瘦素能促进DRG神经元中Bcl-2蛋白的表达,提示瘦素可通过提高Bcl-2蛋白表达来抑制Bax的促凋亡作用,从而改善DRG神经元的存活。这对进一步研究瘦素对DRG功能调节具有重要的意义。
Objective: To investigate the effects of leptin on the expression of Bcl-2 and Bax proteins in cultured neurons of dorsal root ganglion (DRG). Methods: Different concentrations of leptin (1, 10 and 100 nmol/L) were used to treat the cultured DRG neurons. Western blot was used to detect the expression of Bcl-2 and Bax proteins in DRG neurons. Results: 10 and 100 nmol/L leptin could increase the expression of Bcl-2 protein in cultured DRG neurons; However, it could not influence the expression of Bax protein. Conclusion: Leptin can suppress apoptosis of cultured rat DRG neurons through upregulating Bcl-2 expression but inhibiting Bax expression to improve survival of neurons, which should be important to the further research on its physiological function of the DRG.
