爆炸伤创面骨骼肌细胞凋亡的研究 被引量:3

摘要 目的:研究兔肢体爆炸伤创面骨骼肌细胞凋亡、白细胞介素-6(IL-6)和α-肿瘤坏死因子(TNF-α)的表达。方法:新西兰大白兔30只,随机分为炸伤后即刻取材组和炸伤创面普通纱布包扎放置1、3、6、12 h后取材组(n=6);0.9 g铜壳单质猛黑索金炸药(RDX)爆炸装药,海绵间隔4.5 cm,绑于左下肢股部中段前外侧,电引爆;分别取爆炸中心区(Ⅰ区)、边缘区(Ⅱ区)、爆震区(Ⅲ区)炸伤肌肉组织,经原位缺口末端标记法作肌肉组织细胞凋亡检测,ELISA法测定肌肉组织的IL-6、TNF-α含量,并与正常肌肉组织对照研究。结果与结论:爆炸伤后12 h内Ⅰ区偶尔或未检测到凋亡细胞,与正常组织间差异无统计学意义(P>0.05)。Ⅱ区和Ⅲ区肌肉组织在伤后3 h出现凋亡细胞,6 h达到高峰,与正常组织间差异有显著意义(P<0.01);之后逐渐减少,各检测时间点凋亡细胞数Ⅱ区均高于Ⅲ区,差异有显著意义(P<0.01)。炸伤各区组织IL-6、TNF-α含量均高于正常组织含量,差异有显著意义(P<0.01);自Ⅰ、Ⅱ区到Ⅲ区,含量依次降低,差异显著(P<0.01)。Ⅱ区和Ⅲ区中IL-6含量在伤后3 h高表达,6 h达高峰,之后逐渐下降,其变化规律与细胞凋亡情况相似;TNF-α含量在伤后3 h达到高峰,之后逐渐下降。 Objective To study the skeletal muscle apoptosis and the expressions of IL-6, TNF-α of rabbit's limb in the explosive wound. Methods Thirty New Zealand white rabbits were randomly divided into five groups, the first group was taken the tissue from the explosive wound immediately. Other groups were taken the tissue from the wound after explosive wound were bend up for 1, 3, 6, 12 h, respectively(n = 6). 0. 9 g pure euprum strong detonator which had a 4. 5 cm interval with sponge was fastened on the forward and outside of the left limb of the rabbits and ignited with electric impact. The apoptosis of muscular tissue from the explosive center zone ( area Ⅰ ), marginal zone ( area Ⅱ ) and knocking zone( area Ⅲ )was detected through the terminal deoxynucleotidy transferase-mediated dUTP nick labeling; At the same time, to detect the content of IL-6, TNF-α in the muscular tissue through ELISA; And made the control study with the normal muscular tissue. Results and Conclusions The apoptotic cells of area Ⅰ were casually detected or undetected in 12 h after explosive injury, and there were not significant difference between area Ⅰ and normal zone (P 〉 0. 05 ). The apoptotic cells of muscular tissue of area Ⅱ and area Ⅲ started to show up at 3 h after explosive injury, and reached the peak at 6 h, there were significant differences between area Ⅱ and area Ⅲ (P 〈0. 01 ), then fell-off later; the numbers of apoptotic cells in the area Ⅱ were more than those in the area Ⅲ at the each time, and there were significant differences between them(P 〈0. 01 ). The contents of IL-6, TNF-α of each zone were more than those of the norreal tissue, and there were significant differences among them(P 〈0. 01 ) ; and decreased in tum from area Ⅰ to area Ⅲ, which had significant differences among them. The contents of IL-6 in the area Ⅱ and area Ⅲ had high expression at 3 h, and reached the peak at 6 h, then fell-off later, their change regularity was similar to the apoptosis. However, the contents of TNF-α reached the peak at 3 h after explosive injury, then fell- off later
