摘要
目的:检测Galectin-3和骨桥蛋白(OPN)在乳腺浸润性导管癌(IDC)中的表达情况,并探讨其在IDC进展过程中的意义。方法:应用组织芯片技术和免疫组化法研究100例乳腺IDC和35例乳腺良性病变中Galectin-3、OPN的表达情况,并统计分析其与年龄、肿瘤最大直径、组织学分级、TNM分期以及淋巴结转移间关系。结果:Galectin-3在乳腺IDC和良性病变中的表达强度具有明显差异(2χ=47.814,P=0.000),OPN在二者中的表达差异无统计学意义。在IDC组,Galectin-3表达强度与TNM分期、淋巴结转移均呈正相关关系(分别为:rs=0.300,P=0.002;rs=0.335,P=0.001),与年龄、肿瘤最大直径、组织学分级的关系无统计学意义;OPN的表达强度与肿瘤最大直径、TNM分期以及淋巴结转移均呈正相关(分别为:rs=0.236,P=0.018;rs=0.389,P=0.000;rs=0.404,P=0.000),与年龄、组织学分级的关系无统计学意义;Galectin-3与OPN的表达也呈正相关关系(rs=0.266,P=0.007)。结论:Galectin-3在乳腺良、恶性病变中的表达水平具有明显差异,在乳腺病变中检测Galectin-3的表达对病变性质的判断具有重要意义;高表达Galectin-3、OPN的乳腺IDC具有更高侵袭性和淋巴结转移能力,对二者作用机制的深入研究有可能使其成为乳腺IDC患者个体化治疗的新靶点。
Objective To investigate the association between expression of Galeetin-3 and osteopontin (OPN) and clinicopathological parameters in breast cancer patients. Methods lmmunohistochemical assessment of Galectin-3 and OPN expression was performed on a tissue microarray containing 100 cases of breast invasive ductal carcinoma and 35 cases of benign breast lesion. The association of this two-gene expression with clinicopathological parameters ( such as age, T stage, differentiation, TNM stage and lymph node metastasis) was then evaluated. Results The expression in.tensity of Galectin-3 protein was significantly different between the benign and malignant breast lesions (X^2 = 47. 814, P =0. 000). There was no significant difference of OPN expression between benign and malignant mammary tissues. In breast invasive ductal carcinoma, expression of Galectin-3 was significantly associated with TNM stage (rs =0. 300,P = 0.002) and axillary lymph node metastasis( rs =0. 335 ,P =0.001 ) ;Moreover, OPN expression was significantly associated with the tumor size ( rs = 0. 236, P = 0. 018 ), TNM stage ( rs = 0. 389, P = 0. 000 ) and lymph node metastasis ( r, = 0.404,P = 0. 000 ). Furthermore, Galectin- 3 expression has a positive relationship with the expression of OPN (rs =0. 266 ,P = 0. 007 ) in breast invasive ductal carcinoma. Conclusions This study demonstrates different expression intensity of Galectin-3 in benign and malignant breast lesions, which may help in differential diagnosis of breast lesions. High level of Galectin-3 and OPN expression in invasive ductal carcinoma indicate their role in tumor cell invasiveness and metastasis potential. It may be an important and useful strategy to target these molecules for prevention and therapy of breast cancer.
