摘要
目的优化白花丹高效液相色谱法指纹图谱(HPLC-FPs)的分离条件。方法以反相高效液相色谱法,使用Shim-pack VP-ODSC18柱(250mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸水溶液,检测波长270nm,柱温25℃,考察白花丹不同提取物在不同梯度洗脱条件下的分离情况。结果优选出的白花丹HPLC-FPs分离条件可使白花丹不同提取物中各组分达到较佳分离、稳定性、精密度及重复性。结论该色谱分离条件可用于建立白花丹药材HPLC-FPs的分离条件。
Objective To optimize the separation conditions of fingerprint for Plumbago zeylanica by high performance liquid chromatography. Methods The reversed-phase high performance liquid chromato-graphic method was adopted. The samples were extracted by different ethanol (V/V) solution and separated with a linear gradient elution on Shim-pack VP-ODS C la column(250 mm × 4.6 mm, 5 μm) , and then, with the acetonitrile-0.1% phosphoric acid solution as mobile phase, were detected at UV 270 nm and column temperature 25 ℃. During the linear gradient elntion program, the volume fraction of acetonitrile in mobile phase changed as follows: 0 - 80 min, from 15% to 45% ; 80 - 120 min, from 45% to 69% ; 120 125 min, from 69% to 100% ; 125 - 140 min, 100% ; flow rate was 1 mL ·min^-1. Results The separation conditions described above achieved good result in the aspects of separable effect, stability, sensitivity, and reproducibility. Conclusion The optimum separation conditions would be useful for establishing the method of fingerprint for Plumbago zeylanica by high performance liquid chromatography.
