摘要
观察四氢异葎草酮(Tetrahydro-isohumulone,TH)通过线粒体途径诱导SGC-7901细胞凋亡的机制.MTT法检测TH对SGC-7901细胞增殖的影响;荧光显微镜观察TH对SGC-7901细胞凋亡形态的影响;流式细胞仪分析SGC-7901细胞凋亡率;采用试剂盒检测Caspase-9的活性;Western blot法检测Caspase-3蛋白表达情况.2、8、32μg/mL的TH处理SGC-7901细胞72 h后,SGC-7901细胞生长受到抑制,抑制率在一定范围内呈剂量、时间相关性;随着TH质量浓度的提高SGC-7901细胞的凋亡率也增加;TH各剂量均能够显著升高SGC-7901细胞内Caspase-9活性,并呈剂量依赖性;同时,TH能够上调SGC-7901细胞内Caspase-3的表达量,并且呈剂量依赖性.TH可通过诱导SGC-7901细胞凋亡发挥抗肿瘤作用,启动线粒体凋亡通路可能是TH诱导SGC-7901细胞凋亡的重要机制之一.
To investigate the inducing apoptotic effect of Tetrahydro-iso-humulone (TH) on SGC-7901 cells via the mitochondrial pathway. SGC-7901 cells are treated with 2,8,32μg/ mL of TH for 72 hours. The MTT assay is used to detect the effect of TH on SGC-7901 cells in vitro. Fluorescence microscope is used to observe the morphological changes of SGC-7901 cells. Flow cytometry is employed to analyse the apoptotic rates. Activity test kit is used to investigate activity changes of Caspase-9. Western Blot assay is employed to detect the expression of Caspase-3 protein. TH could inhibit the proliferation of SGC-7901 cells in a dose-dependent and time-dependent manner. Apoptotic rates of SGC-7901 cells increased after treated with different dosages of TH, and the activity of Caspase-9 in SGC-7901 is increased by TH in a dose-dependent manner. TH could also up-regulate the expression of Caspase-3 protein in SGC-7901 cells in a dose-dependent manner. TH can inhibit the proliferation of SGC-7901 cells, and inducing cell apoptosis via the mitochondfial pathway may be one of the important action mechanisms of TH.
