莪术油注射液致人卵巢癌SKOV_3细胞胀亡的氧化应激效应 被引量:3

The Effect of Oxidative Stress of Zedoary Turmerie Oil Injection Over Oncosis of Human Ovarian Cancer SKOV_3 Cell

摘要 目的:从氧化应激及线粒体损伤方面探讨莪术油注射液对人卵巢癌SKOV3细胞胀亡可能的作用机制。方法:以莪术油注射液作用人卵巢癌SKOV3细胞后,通过全自动生化分析仪测定细胞上清液乳酸脱氢酶(LDH),荧光染料2',7'-二氯荧光素乙二酯(DCFH-DA)、10-壬基吖啶橙(NAO)结合荧光化学发光仪分别检测细胞内活性氧(ROS)及线粒体心磷脂(MTCL)水平。结果:莪术油注射液作用SKOV3细胞后可使细胞上清液LDH水平上升,细胞内ROS生成增加,MTCL氧化损伤。结论:氧化应激诱导SKOV3细胞线粒体损伤,可能是莪术油注射液致SKOV3细胞胀亡的作用机制之一。 Objective :To discussing the possible oncosis mechanism of zedoary turmerie oil (ZTO) injection over hu- man ovarian cancer SKOV3 from oxidative stress and mitochandrial injury aspects. Methods : After ZTO injection acted on human ovarian cancer SKOV3 cells, to mensurating cell supernatant lactate dehydrogenase ( LDH), fluorescent dyes 2, 7- dichlorofluorescein ethylene ester ( DCFH - DA), 10 - Nonyl acridine orange (NAO) by automatic biochemical ana- lyzer. And detecting the ceils of reactive oxygen species (ROS) and mitochondrial cardiolipin (MTCL) level combined with fluorescent chemiluminescence instrument. Results : After the zedoary oil injection acted on human ovarian cancer SK- OV3 cells, the cell supernatant LDH level riscd, the intracellular ROS generation increased, and got oxidative damage of MTCL. Conclusion: Oxidative stress - induced mitochondrial damage SKOV3 cells, suggesting that their may be one of the mechanisms in ZTO injection over SKOV3 oncosis.
