摘要
采用125I标记示踪结合三氯醋酸(TCA)沉淀法,测定免血浆中表皮生长因子(EGF)的浓度。本方法最小检出量为3.0μg/L,回收率为(97.80±4.94)%,日内变异系数CV为(3.23±2.04)%,血浆在12.5—400μg/L浓度范围内呈良好的线性关系,r=0.9990。本法与酶联免疫吸附分析法(ELISA)对兔血浆样品的测定结果基本一致,但前法测定值略高于后者。
Radioisotope tracer method, combined with trichloroacetic acid (TCA) precipitation, was used to determine the epidermal growth factor (EGF) in plasma of rabbits. The detection limit for EGF in plasma was 3.0 μg/L. The average recovery of EGF was (97.80±4.94)%. The within day-assay coefficient of variation was (3.23±2.04)%. The assay linearity was determined over the range of 12.5- 400μg/L in plasma and the correlation coefficient was 0.9990. The EGF concentration in rabbits was also determined by the ELISA method, the results of these two methods showed little difference on the whole, but present method was slightly greater than ELISA method.
