分泌性中耳炎动物模型鼓室黏膜上皮中转化生长因子β1的表达

The Expression and Significance of Transforming Growth Factor β1 in the Middle Ear Mucosa of Guinea Pigs with Otitis Media With Efussion

摘要 目的检测转化生长因子β1(transforming growthfactor β1,TGF-β1)在分泌性中耳炎(otitis mediawith effusion,OME)动物模型鼓室黏膜上皮中的表达,探讨TGF-β1在OME发病机制中的作用。方法取健康成熟白色豚鼠70只,随机分成7组,每组10只。正常对照组不做任何处理,直接处死提取听泡标本;其余60只为实验组,将灭活肺炎链球菌注入该组豚鼠中耳腔,制作OME动物模型,再将其分为注射后6小时、1天、3天、7天、14天、30天组,采用免疫组织化学方法分别检测各组豚鼠鼓室黏膜上皮TGF-β1的表达。结果在造模成功后的6小时、1天、3天实验组鼓室黏膜上皮中TGF-β1的表达与对照组比较差异无统计学意义,在注射后7天实验组TGF-β1表达明显增强,第30天时达高峰,均明显高于对照组(P<0.05)。结论灭活肺炎链球菌致OME动物模型中,TGF-β1在鼓室黏膜中过度表达,且随病程延长表达增强。 Objective To explore the expression of transforming growth factor β1 (TGF-β1) in the mucosa of guinea pigs with otitis media with effusion(OME) and the role of TGF-β1 in the development of OME. Methods 70 guinea pigs were randomly devided into 7 groups,each with 10 animals. OME was produced by injecting deactivated streptococcus pneumoniae into the typmpanic cavity of guinea pigs. The expression of TGF-β1 was examined by means of immunohistochemistry 6 h, 1 d, 3 d, 7 d, 14 d and 30 d after injection. Results TGF-β1 expression could be detected in the middle ear mucosa at 7 d after injection and reached maximum at 30 d. Conclusion TGF-β1 is expressed in the middle ear mucosa of the guinea pigs with OME induced by deactivated streptococcus pneumoniae, and this indicates that TGF-β1 may play a role in the chronic protraction of OME.
