摘要
[目的]探讨超声-微波辅助技术提取桦褐孔菌多糖的最佳工艺条件。[方法]以水作为提取溶剂,用超声-微波辅助提取桦褐孔菌多糖,通过响应面分析法考察微波功率、微波处理时间和料水比对桦褐孔菌多糖得率和纯度的影响,优化超声-微波提取桦褐孔菌多糖的工艺参数,并和传统水浴浸提法进行比较。[结果]超声-微波辅助技术提取桦褐孔菌多糖的最佳工艺条件为:提取时间18.45~24.50 min,料液比1∶20,微波功率88.3~96.7 W。与传统的水浴浸提法相比,超声-微波提取法可大大缩短提取时间,得率由2.12%增加到3.25%,纯度由64.03%增加到73.16%。[结论]与传统的水浴浸提法相比,超声-微波提取法不仅缩短了提取时间,而且提高了桦褐孔菌多糖的得率和纯度。
[ Objective] The study aimed to discuss the optimum technology condition for the ultrasonic microwave assisted extraction of polysaccharides from Inonotus obliquus. [ Method ] With water as the extracting solvent, the polysaccharides from L obliquus were extracted by ultrasonic microwave assisted extraction technology. The effects of microwave power,microwave treating time and material-liquid ratio on the yield and purity of polysaecharides from L obliquus were investigated through the response surface analysis. The technology parameters for extracting polysaccharides from L obliquus by ultrasonic microwave assisted extraction were optimized and they were compared with that by traditional hot water extraction. [ Result] The optimum technology condition for ultrasonic microwave assisted extraction of polysaccharides from I. obliquus was determined as follows:the extraction time ranged from 18.45 rain to 24.50 min, the material-liquid was 1:20 and the microwave power ranged from 88.3 W to 96.7 W. Compared with the traditional hot water extraction,the ultrasonic microwave assisted extraction could shorten the extraction time greatly and increase the yield from 2.12% to 3.25% as well as the purity from 64.03% to 73.16%. [ Conclusion] Compared with the traditional hot water extraction, the ultrasonic microwave assisted extraction could not only shorten the extraction time greatly, but also increase the yield and purity of polysaccharides from L obliquus.
