摘要
目的观察人绒毛膜促性腺激素(hCG)对滋养层细胞基质金属蛋白酶MMP-19表达的影响。方法用逆转录多聚酶链反应(RT-PCR)检测方法,观察不同浓度hCG于不同处理时间,对来源于人绒癌组织的JEG-3细胞系表达基质金属蛋白酶MMP-19的影响。将JEG-3细胞分别用0.05、0.5、5、25u.mL-1hCG处理48h。结果JEG-3细胞中,MMP-19 mRNA的含量降低;但降低程度与hCG作用浓度的关系不明显。25u.mL-1hCG处理对JEG-3细胞中MMP-19表达的影响有一定的时间效应,以温育30h以上的作用较明显。结论hCG可抑制细胞系JEG-3中MMP-19的表达,并可能通过该机制影响滋养层细胞的侵袭性。
Objective To investigate the effect of human chorionic gonadotrophin (hCG) on matrix metalloproteinase-19 (MMP-19) mRNA expression in trophoblasts.Methods The effect of hCG on MMP-19 mRNA expression in human choriocarcinoma-derived JEG-3 cell line was detected by employing the semi-quantitative method of reverse transcription polymerase chain reaction (RT-PCR). Results After a 48 hours incubation with 0,0.05,0.5,5,25 u·mL^-1 hCG respectively,the expression of MMP-19 in JEG-3 cells decreased,while no difference was found among groups which were treated with different dosages of hCG. Moreover, MMP-19 mRNA expression in JEG-3 cells incubated with 25 u·mL^-1 hCG decreased gradually with the time. Conclusion Human ehorionic gonadotrophin (hCG) might influence the invasiveness of trophoblasts by decreasing the expression of MMP-19.
