摘要
目的:观察机械牵张应变作用下,人牙周膜细胞(HPDLCs)形态及游离Ca2+浓度的变化。方法:通过原代和传代培养获得性状稳定的人牙周膜细胞,使用动态机械应变细胞加载仪对细胞进行1%、10%和20%动态牵张应变加载,并加载不同时间,通过流式细胞仪对细胞进行Ca2+浓度的检测。应用SPSS13.0软件对数据进行方差分析。结果:细胞内游离Ca2+浓度随牵张应变时间的延长逐渐升高,至60min时达到该组的峰值(P<0.01);以后随加载时间的延长逐渐下降,120min时降至对照组水平(P>0.05)。结论:牵张应变可以引起细胞形态的改变和细胞内游离Ca2+浓度的变化。
Objective: The purpose of this study is to investigate the effects of mechanical stretching on morphology and concentration of free calcium in the human periodontal ligament ceils (HPDLCs). Method: Primany human periodontal liga- ment ceils were obtained and passaged. Then the ceils were stretched by dynamic mechanical strain unit with 1%, 10% and 20% strain for different time. Free Ca2+ concentration were examined through Flow cytometry. The data were analyzed by one-way ANOVA using SPSS 13.0 software package. Result: It was significantly higher of the concentration of the free Ca2+ in ceils which were stretched for 60min than that in the control cells (P 〈 0.01) : As the stretching time was prolonged, the concentration of the free Ca2+ decreased to the level of that of control ceils after 120 min stretching. (P 〉 0.05). Conclu- sion: Stretching force not only influenced morphology but also induced the change of concentration of the free Ca2+ of cells.
