摘要
以鱼腥草叶片为外植体,比较不同植物生长调节剂、抗褐化剂和光照条件对愈伤诱导率及褐化率的影响。结果表明,MS+6-BA 0.5 mg.L-1+2,4-D 1.0 mg.L-1有利于鱼腥草愈伤组织的诱导;抗坏血酸(Vc)、聚乙烯吡哆烷酮(PVP)对抑制外植体褐变均有作用,其中,PVP效果最佳,最适浓度为2 g.L-1。光照条件对鱼腥草褐变的影响较大,培养初期暗培养,能显著地抑制外植体的褐变。
In this paper,we used the leaves of Houttuynia cordata Thunb as experiment material to compare the effect of different hormones,anti-browning agents and light on callus inducing and browning.The results indicated that MS addition with 6-BA 0.5 mg·L^-1 and 2,4-D 1.0 mg·L^-1 was the optimal induction medium.VC and PVP can restrained browning of the explants.The effect of PVP was the best,its optimal concentration was 2 g·L^-1.Light had obvious effect on browning of Houttuynia cordata Thunb.Browning of the explants could be restrained obviously without light at the early time of tissue culture.
