睾丸炎动物模型的建立(二)——组织化学和免疫组织化学研究 被引量:2

ESTABLISHMENT OF ORCHIDITIS MODEL IN KM MICE (Ⅱ): HISTOCHEMICAL AND IMMUNO HISTOCHEMICAL STUDY

摘要 目的:研究小鼠睾丸间质细胞和睾丸内细胞外基质在实验性睾丸炎期间的变化。方法:应用组织化学和免疫组织化学的方法分别显示正常小鼠和模型小鼠术后各周睾丸中脂滴、一系列酶及胞外基质的分布和变化。结果:间质细胞胞质内脂滴在实验组术后2、3周明显增加,术后6周有所恢复;非特异性酯酶、3β羟甾脱氢酶、琥珀酸脱氢酶活性在术后1、2、3周减弱,术后5、6周有所恢复;纤维粘连蛋白、层粘连蛋白、Ⅳ型胶原在术后1、2、3周表达量减少,术后6周略有回升。结论:发生睾丸炎期间,睾丸间质细胞功能下降,睾丸内细胞外基质受损。 Objective: To observe the change of Leydig cell and extra cellular matrix (ECM) of testis in mice during experimental orchiditis. Methods: Histochemical methods were used to study the changes of nonspesterases (Nse), 3β hydroxysteroid dehydrogenase (3β HSDH), succinate dehydrogenase (SDH) in Leydig cell and immunohistochemical method were used to observe the changes of fibronectin (FN), laminin (LN), type Ⅳ collagen (Ⅳ C) in ECM between the normal mice and the model mice respectively. Results: 2 and 3 weeks after the operation, the amounts of lipid in Leydig cells of the experimental groups increased, and decreased after the 5th week. The activity of Nse、3β HSDH、SDH was decreased from the 1st week to the 3rd week, and increased slightly after the 6th week but did not reach the normal level. The expression of FN, LN and Ⅳ C was the same as that of the enzyme mentioned above. Conclusion: The decreased function of Leydig cells and damage of ECM in testis during experimental orchiditis might be the factors associated with the poor fertile ability.
