摘要
目的:探讨六亚甲基二乙酰胺(HMBA)联合辐射对人粘液表皮样癌MEC-1细胞超微结构的作用特点。方法:实验分4组,对照组细胞未经药物及射线处理;HMBA组细胞经1mmol/L的HMBA诱导作用96h;γ-射线组细胞在乏氧状态下接受7Gy的60Coγ-射线照射;联合应用组细胞按上述方法先经HMBA处理再接受辐射,用扫描电镜和透射电镜观察各组细胞的形态变化。结果:HMBA组细胞表面微绒毛减少,胞浆线粒体增多;γ-射线组细胞微绒毛凝聚,线粒体肿胀、空化;联合应用组细胞损伤明显,微绒毛凝聚、崩解,线粒体粘连、缺嵴、空化和紊乱。结论:HMBA能增强γ-射线对MEC-1细胞超微结构的破坏作用。
Objective: To investigate the effects of hexamethlene bisacetamide(HMBA) combined with γ-ray on the ultrastructure of human mucoepidermoid carcinoma MEC-1 cell line. Methods: MEC- 1 cells were treated with 1 mmol/L of HMBA for 96 h, 7 Gy of γ ray under anaerobic condition, or their combination respectively and then cells were observed under the scanning elctron microscopy and the transmission elctron microscopy. Results: Reduction of microvilli on the cell surface and increase of mitochondria in the cytoplasma were observed in HMBA treated cells; microvilli coagulation in γ ray treated; severe destruction of microvilli and mitochondria in HMBA and γ ray treated.Conclusion: HMBA can enhance the destructive effects of γ ray on ultrastructure of MEC-1 cells.
